Jordan Peele è nato a New York ed è stato cresciuto dalla madre single nell'Upper West Side di Manhattan. Si è diplomato alla Calhoun School nell'Upper West Side di Manhattan e ha frequentato il Sarah Lawrence College. Dopo due anni, Peele ha abbandonato gli studi per formare un duo comico con la sua compagna di stanza al college e futura autrice di Key & Peele, Rebecca Drysdale. Dal 2016 è sposato con la comica Chelsea Peretti. Nel 2018 ha vinto il suo primo Oscar per aver scritto il suo debutto alla regia, Get Out.
